农杆菌介导的沙田柚基因转化系统建立研究

Taking Epicotyls of Shaddock as the testing objective, a research on effecting factors of establishment of gene transformation system of agricultural bacillus media was conducted. The result of the research indicated that the selecting concentration of Chanamycin of Epicotyl of Shaddock was a little bit over 70mg,that the input of As/150uM into the common culture medium of Epicotyls and agricultural bacillus could increase Epicotyls Gus, which delivered positive rate promptly,and that a preferential duration for seedling was 20-25 days, the preferential cultivating duration for Epicotyls 3 days. At the beginning of selecting Chananmycin, a litter bit over 50mg was in favor of the increase of transformation rate.
